Síntese do Projeto: Hypoglycaemia is a common cause of morbidity in patients treated with insulin, such as those with diabetes. It results from the failure of counter-regulatory mechanisms, particularly the impaired secretion of glucagon, the principal glucose-elevating hormone released by pancreatic α-cells. However, the mechanisms underlying the development of these defects remain unknown.
In this study, we will apply a comprehensive set of advanced techniques—including electrophysiology, live-cell imaging, biochemical assays, and transcriptomic analysis—to dissect a novel regulatory pathway of glucagon secretion and investigate how it becomes disrupted under pathological conditions.
Ultimately, our goal is to identify strategies to prevent recurrent hypoglycaemia and establish mechanistic foundations for the development of therapeutic interventions that could significantly improve diabetes management.
