Objetivo temático: Overall, with this project we intend to characterize how specific intracellular AMPK recruitment affects MERCs’ dynamics and subsequent biological functions, including metabolism. These mechanisms are already recognized as being dysregulated in several age-related pathologies, and their characterization will probably expose novel therapeutic targets to tackle them.

Síntese do Projeto: How the communication between mitochondria and the endoplasmic reticulum (ER) is modulated by the recruitment of specific intracellular AMPK intracellular pools. Intracellular AMPK compartmentalization probably occurs in order to precisely regulate a coordinated downstream response to specific stimuli, although the regulating mechanisms remain obscure. Based on a robust set of preliminary data, we have compelling evidence that AMPK signaling modulates mitochondria-ER contacts, which mays have functional consequences during physiological and pathological conditions. By precisely modulating AMPK activity, allied to a complete set of functional outcomes at both mitochondrial and mitochondria-ER contact levels, we will contribute to the development of an integrated cellular landscape linking specific AMPK-mediated metabolic effects, mitochondria-ER crosstalk, and mitochondrial physiology.
